you can use tablets or you can use the pencil/brush/pen tools. you can also scan in lineart.

You could use the vector pen and redraw your line over the top of your image *Link Removed*(PM me if some mod deletes the link) has some great tutorials for this technique. Click on Drawing Tutorials at the top, then choose which option you need under "computer colouring" in the left hand side bar.

I personally use custom select, all black areas that i want, and after this torture test...I go "EDIT" , "Fill" , and tada ^^... but u can just use Stroke and get all selected border painted.
