Kujibiki Unbalance episode 2, karaoke scene. A damn great one as well.
There are of course many more, but those two are the ones I did remember first and also watched several times just for the heck of it. :)
NHK ni Youkoso! for the most funniest manga, as really every scene had me laughing till I shed tears. Luckily an anime based on it will air next year in Japan. :D
Other pretty funny anime/manga: Azumanga Daioh, Yotsubato, Genshiken, Full Metal Panic Fumoffu, Soldiers of Wishful Thinking. Love Hina can be considered somewhat funny as well I guess.

i remember on one slam dunk episode when sakuragi slam dunked takenori akagi's head on there silly one on one, men! i laughed so hard on that episode
every minute was hilarious
